早期流产大鼠子宫中IL-18和NF-κB及iNOS的表达

摘要:
[目的]检测了早期流产、正常妊娠和非妊娠大鼠子宫中白细胞介素-18(Interleukin-18,IL-18)、核转录因子(Nuclear factor-kappa B,NF-кB)及诱导型一氧化氮合酶(Inducible nitric oxide synthase,iNOS)的分布与表达,探讨IL-18、iNOS和NF-кB的表达与流产的关系.[方法]采用阴道涂片法将经过自然交配的大鼠分为流产组、正常妊娠组和非妊娠组3组,每组10只.流产组大鼠于妊娠第7天和第8天分别灌服米非司酮4 mg/(kg·d).各实验组大鼠均于交配后第9天处死,取子宫,切片,免疫组织化学SP法染色后进行图像分析.[结果]IL-18、NF-кB和iNOS在正常妊娠组、非妊娠组和流产组大鼠子宫腔上皮细胞、腺上皮细胞、子宫基质细胞或蜕膜细胞、子宫肌层平滑肌细胞及部分血管内皮细胞中均有表达.与正常妊娠组相比,流产组大鼠子宫中IL-18和iNOS的表达极显著减弱(P<0.01),NF-κB表达极显著增强(P<0.01),血管内皮细胞和平滑肌细胞中IL-18、NF-кB和iNOS的表达均极显著增加(P<0.01).[结论]IL-18、NF-кB和iNOS的异常表达可能与流产有关.

Expression of IL-18, NF-кB and iNOS in early abortion rat uterus
Abstract:
【Objective】 The research was to study the distribution and expression of IL-18,NF-кB and iNOS in early abortion,early pregnancy and nonpregnancy rat uterus and to understand the possible relation between these factors and abortion. 【Method】 The natural mating rats were divided into three groups (nonpregnancy group,pregnancy group and abortion group)by observing vagina secretion with ecah group of 10 rats.The abortion group rats were perfused mifeprestone,4 mg/(kg·d),in the pregnancy on day 7 and 8.All rats were killed on day 9.The rats uterus were sliced up and done by immunohistochemical technique and image analysis.【Result】 IL-18,NF-кB and iNOS protein can be detected in all rats uterine endometrial or decidual luminal epithelium,glandular epithelium,stroma or decidual cells,myometrium and part endothelial cell and vascular smooth muscle cell.Compared with normal pregnancy group,the expression of IL-18 and iNOS in abortion rats was distinctly decreased (P<0.01);The expression of NF-кB in abortion rats was distinctly increased (P<0.01).However,the expression of IL-18,iNOS and NF-кB in abortion rats endothelial cell and vascular smooth muscle cell was markedly increased (P<0.01). 【Conclusion】 It can be concluded that the abnormal expression of IL-18 ,iNOS and NF-кB may have relation with abortion.
