人卵泡液对牛未成熟卵母细胞体外培养的影响

摘要:
[目的]探讨以人成熟卵泡液作为体外成熟培养液培养牛卵母细胞的可能性.[方法]观察并比较未成熟牛卵母细胞分别在灭活人卵泡液与成熟培养液、灭活与未灭活人卵泡液、未灭活人卵泡液不同培养温度(38,38.5和39℃)下,进行体外成熟培养时卵母细胞的极体排出率.[结果]在灭活人卵泡液与成熟培养液中,牛卵母细胞成熟率分别为58.5%和63.5%,二者无显著性差异(P>0.05);在未灭活人卵泡液中,牛卵母细胞成熟率(58.8%)显著高于灭活人卵泡液(37.5%)(P<0.01);在未灭活人卵泡液中38,38.5和39℃培养的牛卵母细胞的成熟率分别为60.7%,58.1%和54.1%,三组间差异不显著(P>0.05).[结论]人卵泡液可以用于未成熟牛卵母细胞体外成熟;人卵泡液用作成熟培养液时无需灭活,培养温度为38~39℃.

Effects of human follicular fluid on in vitro maturation rate of bovine oocytes
Abstract:
【Objective】 The study was to evaluate the effect of human follicular fluid (HFF) on bovine oocytes matured in vitro.【Method】 In this experiment,immature bovine oocytes were cultured in HFF and in vitro maturation (IVM) medium,inactivated HFF and uninactivated HFF,inactivated HFF under different temperatures (38,38.5 and 39 ℃).The rates of maturating were compared.【Result】 The rates of maturating in the HFF group (58.5%) was lower than that in the IVM medium group (63.5%),but their differences were not significant (P>0.05).The rates of maturing in the inactivated HFF group (37.5%) was significantly lower than that in the group (58.8%) which was not inactivated (P<0.01).There were also no significant differences among the rates of maturing in the three groups (38,38.5 and 39 ℃) (P>0.05).Their rates of maturing were 60.7%,58.1% and 54.1% respectively.【Conclusion】 Human follicular fluid is effective in the bovine oocytes matured in vitro.Uninactivated HFF is more effective for culturing bovine oocytes.When using uninactivated HFF,the culture temperature shoule be among 38-39 ℃.
