固原本地黄牛及其利杂群体Pit-1基因多态性与育肥性状的关系

摘要:
为了分析固原本地黄牛及其利木赞牛杂交群体(简称利杂群体)Pit-1基因的多态性与育肥性状的关系,为固原本地黄牛下一步的杂交改良提供理论基础,利用PCR-RFLP技术研究了固原本地黄牛及其利杂群体共178个个体的Pit-1基因多态性,并分析了其与育肥个体育肥性状的相关性。结果表明,451 bp的PCR产物被限制性内切酶HinfⅠ消化后表现出多态性,固原本地黄牛及其利杂群体在该位点均处于Hardy-Weinberg平衡状态,等位基因A/B的频率分别为0.303 9/0.696 1和0.161 4/0.838 6。与部分育肥性状的相关分析表明,在育肥的初期阶段,BB型和AB型个体的精料料肉比均显著低于AA型个体(P<0.05);7月龄育肥牛BB型个体的胸深显著低于AA型,而胸宽显著高于AA型(P<0.05);育肥牛6~12月龄体长增长量也表现出BB型个体优于AB型的趋势(P<0.05)。初步认为,BB基因型与优良的育肥性能显著相关,对B等位基因的选择有利于优良性状的改良。

Association of polymorphism of Pit-1 gene with fattening traits
Abstract:
PCR-RFLP was applied to analyze the polymorphisms of Pit-1 gene in 178 Guyuan local cattle and crossbreed of Limosin×local cattle,and then the association with fattening traits had been analyzed.The results showed that 451 bp PCR products digested with HinfⅠ demonstrated polymorphisms in Guyuan local cattle and crossbreed of Limosin×local cattle which were all at Hardy Weinberg equilibrium,and the frequencies of allele A/B were 0.303 9/0.696 1 and 0.161 4/0.838 6,respectively.The association of polymorphisms of Pit-1 gene with some fattening traits showed that the individuals with genotype BB and AB had lower concentrates to gain ratio in the first stage of the fattening than the individuals with genotype AA (P<0.05);the individuals with genotype BB had bigger chest width and smaller chest depth than the individuals with genotype AB (P<0.05),the individuals with genotype BB had better tendency in body length increment in 6-12 months than the individuals with genotype AB(P<0.05).So the genotype BB was significantly associated with the fine fattening performance and the selection of the allele B was better for the trait improvement.
