小鼠卵巢颗粒细胞的体外培养

摘要:
为了探讨小鼠卵巢颗粒细胞体外生长的特点及增殖的调控因素,分别添加2 μg/mL胰岛素、50 ng/mL FSH和20 ng/mL EGF的培养液培养小鼠卵巢颗粒细胞,每隔24h对细胞进行计数,研究其对小鼠卵巢颗粒细胞增殖的影响。此外,还对培养过程中出现的卵巢颗粒细胞集落进行了AKP染色和C-kit免疫组化染色鉴定。结果显示,在培养的第4天添加EGF组和FSH组与对照组相比均具有显著的促增殖作用(P<0.05),添加胰岛素组促增殖作用不显著(P>0.05);卵巢颗粒细胞集落经AKP染色和C-kit免疫组化染色均呈阳性。提示EGF和FSH对小鼠卵巢颗粒细胞具有促增殖效应,小鼠卵巢颗粒细胞具有干细胞的部分特性。

Culture of mouse granulosa cells in vitro
Abstract:
To investigate the growth characteristics and regulational factors contributed to proliferation of mouse granulosa cells in vitro,insulin (2 μg/mL),FSH (50 ng/mL) and EGF (20 ng/mL) were added into the basal medium,respectively.The cell number were counted every 24 hours,the effect of each factor on promoting mouse granulosa proliferation was researched.In addition,alkaline phosphatase (AKP) staining,and C-kit immunohistochemical staining were performed to identify the clones of granulosa cells appearing in culture.The results showed that the group added with EGF and FSH,but insulin (P>0.05),had significant effect on promoting proliferation(P<0.05) compared with the control group on the 4th day;the clones of granulosa cells were both positive for AKP and C-kit.Therefore,it was concluded that EGF and FSH could promote granulosa cells’proliferation and mouse granulosa cells had some properties of stem cells.
