冬青卫矛内生真菌2QR1菌株代谢产物的杀菌活性

摘要:
为寻找天然植物源新的具有农药活性的先导化合物,从冬青卫矛根皮中分离到1株内生真菌,编号为2QR1,初步鉴定其为半知菌亚门,曲霉属(Aspergillus)。室内活性测定结果表明,2QR1菌株发酵液的杀菌活性远高于菌丝体的杀菌活性。该菌株发酵液乙酸乙酯萃取物对小麦根腐病菌、小麦赤霉病菌、苹果炭疽病菌、玉米大斑病菌、番茄叶霉病菌、烟草赤星病菌和马铃薯干腐病菌菌丝生长抑制的有效中质量浓度EC50值分别为53.6,63.1,63.3,98.2,106.5,115.9和166.3μg/mL;对玉米大斑病菌、小麦根腐病菌、烟草赤星病菌和马铃薯干腐病菌抑制孢子萌发的有效中质量浓度EC50值分别为94.8,139.1,146.6和180.1 μg/mL;在质量浓度为500 μg/mL时,对枯草芽孢杆菌、蜡状芽孢杆菌和金黄色葡萄球菌的抑菌圈直径在16 mm以上。盆栽试验结果表明,2QR1菌株发酵液乙酸乙酯萃取物在质量浓度为2 000 μg/mL时,对小麦白粉病的保护作用和治疗作用分别为61.9%和81.6%。选用硅胶柱层析,以杀菌活性追踪为指导,对该菌株发酵液乙酸乙酯萃取物进行了杀菌活性成分的分离,得到化合物B9。化合物B9对玉米大斑病菌、烟草赤星病菌、小麦根腐病菌和马铃薯干腐病菌抑制孢子萌发的有效中质量浓度EC50值分别为0.8,1.4,3.3和5.0 μg/mL。

Antifungal metabolites of endophytic fungus of 2QR1 strain in Euonymus japonicus
Abstract:
A endophytic fungi strain was isolated from Everyreen euonymus.The strain numbered 2QR1 was preliminarily identified as deuteromycotina Aspergillum.The result of fungicidal activities of metabolities of 2QR1 strain showed that the EC50 value of ethyl acetate extract of fermentation broth of 2QR1 strain was separately 53.6,63.1,63.3,98.2,106.5,115.9 and 166.3 μg/mL against Bipolaris sorokiniana,Fusarium graminearum,Glomerella cingulata,Exserohilum turcicum,Fulvia fulvum,Alternaria longipe and Fusarium solani.The EC50 value of acetate extract of fermentation broth of 2QR1 to inhibit the spore germinations of Exserohilum turcicum,Alternaria longipes,Bipolaris sorokiniana and Fusarium solani was separately 94.8,139.1,146.6 and 180.1 μg/mL.The inhabition zone diameter of ethyl acetate extract of fermentation broth of 2QR1 strain was more than 16mm at the concentration of 500 μg/mL against Bacillus subtilis,Bacillus cereus and Staphylococcu aureus.The results of pot test indicated that ethyl acetate extract of fermentation broth of 2QR1 strain showed 61.9% of protected efficacy and 81.6% of therapeutic efficacy at 2 000 μg/mL.Compound B9 was separated from the ethyl acetate extract of fermentation broth by p-TLC.The result of spore germination proved that the EC50 value of compound B9 was separately 0.8,1.4,3.3和5.0 μg/mL,against Exserohilum turcicum,Alternaria longipes,Bipolaris sorokiniana and Fusarium solani.
