中国樱桃的PCR-RFLP分析

摘要:
采用三因素五水平正交组合设计,探讨了野生中国樱桃的PCR-RFLP扩增条件及限制性内切酶酶切反应体系,并用PCR-RFLP技术对中国樱桃进行了分子亲缘地理学研究。结果表明,利用42种引物/酶组合,在13个中国樱桃居群中检测出多态位点比率为99.48%;在所有酶切片段中,发现了5种cpDNA单倍型(H1、H2、H3、H4、H5);根据单倍型的地理分布格局,将所研究的中国樱桃居群划分为4个地理单元,地理单元间的基因分化系数(GST)值为0.397,cpDNA基因流(Nm)为0.758;中国樱桃cpDNA总的遗传多样性(Ht)为0.166,地理单元内的cpD-NA遗传多样性(Hs)为0.005。分析结果说明,当第四纪冰川来临时,中国樱桃向南迁移并避难于云南省东南部山区、秦岭南坡及大巴山的东部等地区;当冰期结束、气候缓和时,避难于云南省东南部山区的中国樱桃分别向东北、西北方向回迁,避难于大巴山东段的中国樱桃则向东北方向迁移,形成现今的分布格局。

PCR-RFLP analysis of Prunus pseudocerasus
Abstract:
The PCR-RFLP amplification system and the endonucleation system of Prunus pseudocerasus were studied by using a three-factor orthogonal combination design.Molecular phylogeographical research upon P.pseudocerasus in 13 populations has been carried out by means of the PCR-RFLP reaction system.The percentage of polymorphic loci which was 99.48% was detected by 42 pairs of primer/enzyme.Five cpDNA haplotypes were detected through the analysis of all the fragments.The investigated P.pseudocerasus populations could be divided into 4 geographical units according to the haplotypes.The variation coefficient(GST) was 0.397 and the cpDNA gene flow was 0.758 among geographical units.Total cpDNA genetic diversity was 0.166,and 0.005 in geographical units.The results showed that P.pseudocerasus migrated southwards and inhabited the mountainous areas of southeast Yunnan Province,Qinling's south slope and Dabashan's east during the glacier of Quaternary period.With the alleviation of climate after the end of glacial age,P.pseudocerasus individuals from southeast in Yunnan migrated northwest and northeast respectively while those from Dabashan's east section migrated northeast,and therefore the current distribution pattern was eventually formed.
