培养基中主要成分对酵母生长及富硒性能的影响

摘要:
以活性干酵母为菌种,采用摇瓶培养,通过单因素试验和正交试验设计,研究了发酵培养基中酵母粉、蛋白胨和蔗糖3种主要成分对酵母生物量及富硒性能的影响。结果表明,对酵母生物量影响最主要的营养成分是蔗糖,其次是酵母粉和蛋白胨;对酵母硒含量和硒总含量影响最主要的成分均为蛋白胨,其次为酵母粉和蔗糖。以酵母中硒总含量为指标,初步确定的富硒酵母优化培养基配方为:酵母粉15 g/L,蛋白胨10 g/L,蔗糖20 g/L,且在该营养条件下制得富硒酵母的硒含量和硒总含量分别可达1 050.00 μg/g和5 510.07μg/L,与优化前(培养基配方为酵母粉10 g/L,蛋白胨10 g/L,蔗糖40 g/L)相比分别提高了28.8%和9.01%。

Influence of fermentation medium on the biomass and selenium content of active dry yeast
Abstract:
By using single factor tests and the orthogonal design,biomass and Se content of yeast influenced by three factors:yeast extract powder,peptone and sucrose of fermentation medium,were studied.The results showed that sucrose was the most principal factor in the three factors that affected the yeast biomass,and peptone was the more primary factor that affected the Se content and the total Se content of yeast than yeast extract powder and sucrose.The optimum fermentation medium was determined as:yeast extract powder 15 g/L,peptone 10 g/L,and sucrose 20 g/L by index of the total Se content.Under the initial optimized fermentation medium,Se content and the total Se content of selenium enriched yeast reached 1 050.00 μg/g and 5 510.07 μg/L,and improved by 28.8%,and 9.01% respectively,compared with that of the previous ingredient (yeast extract powder 10 g/L,peptone 10 g/L,and sucrose 40 g/L) of culture medium.
