小麦幼穗的离体培养及其影响因素研究

摘要:
为了确立适于小麦幼穗遗传转化的组织培养体系,对8个小麦品种的幼穗通过组织培养方法,研究了不同基因型、培养基对愈伤组织诱导及分化的影响。结果表明,不同小麦品种幼穗愈伤组织诱导出愈率差异不大,而分化率却差异明显,其中绵阳19分化率最高,平均可达72.9%,洛阳8716次之,平均为61.2%;分化培养基对分化率也有很大影响,在选用的2种分化培养基中,郑麦9023幼穗愈伤组织的分化率差异最大,而洛阳8716的差异最小;小偃22、西农88、郑麦9023和陕150的幼穗愈伤组织,在分化培养基B中分化率较高,而陕354、绵阳19和小偃107则在分化培养基A中较高;幼穗愈伤组织分化最佳温度为20~22℃,光照强度3000lx。因此,在所选用的8个小麦栽培品种中,绵阳19和洛阳8716的幼穗愈伤组织可作为遗传转化的受体材料。

Influencing factors on wheat immature inflorescences culture
Abstract:
In order to establish a high frequency of plant regeneration system,the immature inflorescences of eight wheat varieties were cultured to study the effects on the frequency of callus induction and plant regeneration.The result showed that there was little difference between the frequences of callus induced among all varieties,but the frequencies of plantlet differentiation were quite different.Mianyang No.19 was the best genotype for explant regeneration,and Luoyang No.8716 took the second place.The differentiation medium also had great influence on differentiation of different wheat varieties;The optimum differentiation medium of Xiaoyan No.22,Xinong No.88,Zhengmai No.9023 and Shan No.150 was medium B,while Shan No.354,Mianyang No.19 and Xiaoyan No.107 was medium A.The optimum culture condition was temperature 20-22 ℃ and light intensity 3 000 lx.Therefore the immature inflorescences of Mianyang No.19 and Luoyang No.8716 were the optimal explant for wheat transformation.
