木聚糖酶固态发酵培养基的优化

摘要:
采用单因素试验和正交试验对宇佐美曲霉(Aspergill ususamii)YW-38菌株产木聚糖酶的固态发酵工艺条件进行了研究。结果表明,三角瓶优化培养基及发酵条件为:250 mL三角瓶装8.0 g基料(m(麸皮)∶m(玉米芯)=3.5∶4.5),NH4NO3 10g/kg,Tween-80 3 g/kg,KH2PO4 3.0 g/kg,CaCl2 1.0 g/kg,MgSO4·7H2O 1.0 g/kg(均相对于基料),基料与自来水的质量比为1∶1.3~1∶1.4,pH自然;于29 ℃培养72 h,期间翻曲2次,此工艺条件下最高比酶活力为8752IU/g。曲盘发酵工艺条件为:基料与自来水的质量比1∶1.8,其余成分同三角瓶优化培养基,29 ℃培养68 h,比酶活力可达8 761 IU/g。

Optimization of solid-state fermentation medium for Xylanases
Abstract:
The solid-state fermentation for xylanase production by Aspergillus usamii was investigated in this paper.Multivariate statistical approaches were employed to evaluate the effects of several variables (carbon and nitrogen sources,the ratio of base material to water,initial pH and temperature) on xylanase production.Under the optimized process conditions,the xylanase activity reached 8 752 IU/g dry koji cultured with flask at 29 ℃ for 72 h and 8 761 IU/g dry koji with koji tray at 29 ℃ for 68 h.
