以中国西门塔尔牛、安格斯牛和海福特牛为试验材料,设计特异性引物扩增解耦联蛋白3基因(UCP3)的第5内含子和第6外显子部分序列,利用PCR-SSCP技术对牛UCP3基因进行了SNPs检测和基因型分析。结果表明,3个牛品种中的优势等位基因均为B,中国西门塔尔牛和安格斯牛的优势基因型为BB型,海福特牛的优势基因型为AB型;中国西门塔尔牛和海福特牛在UCP3基因位点处于Hardy-Weinberg平衡状态,而安格斯牛在UCP3基因位点处于Hardy-Weinberg非平衡状态;中国西门塔尔牛和海福特牛在UCP3基因位点为中度多态,安格斯牛在UCP3基因位点为高度多态。 |

Study on polymorphisms of part sequence of intron 5 and exon 6 of bovine UCP3 gene |

Abstract: |
Take Chinese Simmental,Angus and Heiford as experiment materials,a pair of primers for the part of intron 5 and exon 6 of uncoupling protein 3 gene were designed.The polymorphisms were analyzed by PCR-SSCP and DNA sequencing.The results showed that allele B was the predominance allele in these three populations,genotype AA was the predominance genotype in Chinese Simmental and Angus and it was AB in Heiford.There was no significant difference (P>0.05) in UCP3 locus of both Chinese Simmental and Heiford,while significant differences existed in Angus.Analysis of polymorphisms index showed the polymorphism of the locus was moderate in Chinese Simmental and Heiford and high in Angus. |
