玉米幼芽提取物对小鼠皮肤成纤维细胞体外抗氧化能力的影响

摘要:
分离培养小鼠皮肤成纤维细胞,将传代细胞均匀分成4组:Ⅰ组为空白对照组;Ⅱ组在培养体系中加入终浓度为200μmol/L的H2O2作用2h,复制氧化损伤模型;Ⅲ组和Ⅳ组分别用含20 mg/L的0501和0502号玉米幼芽提取物培养液培养24 h,然后再用Ⅱ组的处理方法对细胞进行氧化损伤,研究玉米幼芽提取物对皮肤成纤维细胞体外抗氧化效应的影响。结果表明,Ⅲ组和Ⅳ组细胞凋亡率极显著低于Ⅱ组(P<0.01);扫描电镜下Ⅱ组细胞的细胞膜出现皱缩穿孔等现象,Ⅲ组和Ⅳ组细胞完整性较好,细胞联系紧密;透射电镜观察,Ⅱ组细胞粗面内质网扩张,染色质边集,细胞内出现大量空泡,Ⅲ组和Ⅳ组细胞结构清晰,出现轻度的粗面内质网扩张。表明富含腺嘌呤类衍生物的玉米幼芽提取物对体外培养的小鼠皮肤成纤维细胞具有抗凋亡保护作用,在细胞氧化损伤过程中,对细胞膜蛋白和结构性蛋白有保护作用。

Effect of extracts of maize plumule on mouse derma fibroblast in vitro in the oxidative damage condition
Abstract:
During the study,the mouse derma fibroblast (MDF) was separated from mouse skin and cultured in vitro.The cultured MDF were divided into 4 groups:group Ⅰ was control group;H2O2 (200 μmol/L,2 h) was used to make oxidative damage model on MDF in group Ⅱ;the extracts of maize plumule (20 mg/L) also were used in group Ⅲ and group Ⅳ for 24 h,then the MDF in the two groups were disposed in the same way as group Ⅱ.So the antioxdative damage effect of the extracts of maize plumule on the MDF in vitro was studied.The result showed that the apoptosis ratio of MDF in group Ⅲ and group Ⅳ was significantly lower than that of group Ⅱ.Observed by SEM and TEM,Crimple,cracking happened on cell membrane,or even the structure of the membrane disappeared,the chromatin gathered to the karyotheca,microvillus disappeared and vacuolar degeneration also happened in some cells in group Ⅱ,and in group Ⅲ and group Ⅳ,cells showed almost intact structure,but some areas of cell membrane crimpled,dilation of rough endoplasmic reticulum happened,and the phenomenon that the chromatin gathered to the karyotheca appeared.Above all,extracts of maize plumule rich in adenine ramification could prevent MDF from apoptosis,and protect the structure protein and membrane protein when the oxidative damage happened.
