| 采用ELISA-DSM法和半叶枯斑法,研究了壳寡糖诱导对普通烟(Nicotiana tabacum)体内TMV增殖的影响.结果表明,普通烟经50μg/mL壳寡糖溶液诱导处理后,其对TMV的侵染表达了高水平的系统抗病性,显症推迟4~7 d,平均严重度降低了82.9%.ELISA-DSM法检测结果表明,壳寡糖诱导处理的普通烟植株体内病毒浓度明显降低;在接种后10 d,接种叶片OD值仅为0.400,相当于同期不诱导对照的23.0%;新生叶片OD值为0.190,相当于同期不诱导对照的38.7%.以发病普通烟叶片为接种体,利用半叶法接种心叶烟(Nicotiana glutinosa),结果表明,壳寡糖诱导处理大大减少了枯斑数目;以接种后10 d的壳寡糖处理叶片为接种体,平均半叶枯斑数为9.25,仅为不诱导对照的19.5%;以同期新生叶为接种体,平均半叶枯斑数为0.630,为不诱导对照的25.0%.以上两种方法的检测结果均证明,壳寡糖诱导处理可以有效抑制普通烟体内的TMV增殖.通过对壳寡糖(低分子质量)处理与壳聚糖(高分子质量)处理的比较发现,分子质量的差异不影响其对病毒增殖的抑制效果. |

| Induced resistance to TMV multiplication in tobacco with Chito-oligosaccharides |

| Chito-oligosaccharides are elicitors to systemic induced resistance to viral infection in tobacco (Nicotiana tabacum).The inhibitory effect to TMV multiplication in tobacco plants treated with chito-oligosaccharides was examined by ELISA-DSM and local lesion assays.The results showed high level resistance achieved with 4-7 d delayed latent period and 82.9% suppression of average severity in tobacco plants treated with a solution of chito-oligosaccharides at a concentration of 50 μg/mL.In ELISA-DSM assay chito-oligosaccharides were shown to be able to inhibit multiplication of viruses in infected plants.The OD value in inoculated leaves (10 d after inoculation) from chito oligosaccharides treated plants was 0.400,which was equal to 23.0% of that from untreated control plants,and 0.190 in fresh leaves from treated plants,or 38.7% of that from untreated control plants on the same conditions.In local lesion assay the number of local lesions per half-leaf of N.glutinosa infected by TMV inoculum from detached leaves of tobacco treated with chito oligosaccharides,was largely reduced.For example,in the assay with detached inoculated leaves (10 d after inoculation),the average number of local lesions was 9.25 in chito-oligosaccharides treatment,or 19.5% of that in untreated control,and in the assay with fresh leaves only 0.63 local lesions produced in chito-oligosaccharides treatment,equal to 25.0% of that in untreated control.The results obtained from two assays revealed that treatment with chito-oligosaccharides effectively inhibited TMV multiplication in tobacco.Besides,it was found that molecular weight of compounds did not influence the inhibitory effect contrast to treatments with chito-oligosaccharides (low molecular weight) or with chitosan (high molecular weight). |
