摘要: |
通过对影响RAPD标记稳定性的引物浓度梯度、退火温度梯度、dNTP浓度梯度、Taq酶浓度梯度等外部条件的优化研究,确定了RAPD反应适宜的引物浓度为0.1~0.2 pmol/μL,退火温度为48~52 ℃,dNTP浓度为0.3~0.15 mmol/L及Taq酶浓度为0.3 U/μL,提高了该反应体系中RAPD的稳定性。 |

Abstract: |
Factors that affect the stability of RAPD were optimized such as primer concentration grads,annealing temperature grads,dNTP concentration grads and taq concentration grads.The proper concentration of primer was 0.1-0.2 pmol/μL,the proper annealing temperature 48-52 ℃,the proper concentration of dNTP 0.3-0.15 mmol/L,the proper Taq concentration 0.3 U/μL.and the stability of RAPD was enhanced in this system. |
