| 以凯氏定氮法为基准方法,采用考马斯亮蓝法、Folin-酚法、双缩脲法对从重组大肠杆菌发酵产物中提取的类人胶原蛋白含量进行了测定。结果表明,这种类人胶原蛋白和牛血清白蛋白与明胶的特性存在差异,在考马斯亮蓝法、Folin-酚法、双缩脲法3种比色分析方法中,采用双缩脲法得出的类人胶原蛋白与牛血清白蛋白的回归方程系数相差最小,并得出回归方程的校正系数α和β,其中α表示类人胶原蛋白与牛血清白蛋白的特性差异(α=0.676 1),β为类人胶原蛋白样品中蛋白的百分含量(g/g)。 |

| Differences among three methods of determining one kind of human-like collagen |

| Abstract: |
| One kind of human-like collagen was determined in this paper.On the basis of Kjeldahl,Coomassie Brilliant Blue,Folin-hydroxybenzene,Biuret was used to determine the human-like collagen produced from the recombined E.coli after high-density cultivation and purification.The results showed that the property of human-like collagen was different from that of bovine serum albumin (BSA) and gelatin,and the linear coefficient of human-like collagen was close to that of BSA when analyzed with Buiret,coefficient α and β could be made out and the values were 0.676 1 meaning the difference between human-like collagen and BSA and protein percent concentration (g/g) respectively. |
