苹果渣酶促糖化效应的研究

摘要:
以不同的酶及酶组合处理苹果渣,研究了苹果渣中多糖物质转化为可溶性糖的条件与规律。结果表明,淀粉酶单独作用时,60 ℃下苹果渣中淀粉分解最快,只需2.75 h。酶用量在10 g/kg以下时,复合果胶酶对苹果渣的糖化能力最强,总糖生成量达到相同用量纯果胶酶的2倍以上;对不同酶组合处理,在50 ℃条件下,以复合果胶酶取代纤维素酶和果胶酶作用2 h,再加淀粉酶作用4 h的双酶2步法为糖生成量高、又省酶的最优酶促糖化工艺,并且糖生成量随双酶用量的增大而增高,至酶用量3~4 g/kg苹果渣的含糖量提高3~4倍,之后升幅趋于平缓。

Study on the saccharifying effect by enzymolysis with apple pomace
Abstract:
In order to utilize the carbohydrate in apple pomace (AP) effectively and use AP as the valuable raw material of sugar production,ethanol fermentation etc,amylase,pectinase and cellulolase were applied in apple pomace respectively or with different combination of each to find out the necessary condition and law of its polysaccharide turning into soluble sugar.The results showed that 60 ℃ is the optimum temperature of enzymolysis with amylase,under which enzymolysis took only 2.75 h.It was found from experiments of different dosage of different enzymes that compound pectinase might possess more stronger saccharifying ability to AP than other enzymes.The net increasing value of both reducing sugar and total sugar with compound pectinase would be twice that of pure pectinase of same dosage.Among all the treatments in which 3 enzymes were all employed,two steps with double enzymes were selected to be the best effective and lest costly enzymolysis method,that is to compond pectinase/2 h·50 ℃,followed by amylase/4 h·50 ℃,which reducing sugar and total sugar yield would increase with the increase of double enzyme dosage till 3-4 g/kg dosages were arrived,and sugars content were increased by 3-4 times that of untreated pomace.
