普通小麦基因转化中良好受体系统的建立

摘要:
对影响小麦幼胚愈伤组织高频率再分化的条件进行了研究。结果表明,暗培养条件下诱导的小麦幼胚愈伤组织的再分化频率显著高于散光和2 000 lx光照条件;大田生长的小麦比温室盆栽小麦的幼胚脱分化和再分化频率高。小麦主茎穗部的幼胚组织脱分化和再分化特性显著高于小麦分蘖茎穗部的幼胚组织。不同年份、不同基因型小麦品种,及不同胚龄幼胚组织的脱分化和再分化特性有一定差异。通过诸多因素的优化和最适幼胚组织的选择,可建立小麦基因转化中不依赖于基因型的良好受体系统。

Establishment of transgenic acceptor system in wheat (Triticum aestivum L.)
Abstract:
Effect of few factors on wheat immature embryo callu redifferentiation ability were studied.The resulted showed that the redifferentiation frequency of wheat immature embryo callus inducted in dark culture was higher than that of wheat immature embryo callus inducted in nature light and 2 000 lx light.The callus redifferentiation frequency of immature embryos obtained from field was higher than that of immature embryos obtained from greenhouse.The callus redifferentiation frequency of immature embryos obtained from main stem was higher than that of immature embryos obtained from tiller.Different genotype varieties and different development stage embryos were different in the redifferentiation characteristic of immature embryo callus.Wheat transgenic acceptor system has been established using optimization of culture factor and dedifferentiation culture of wheat immature embryo.
