对小麦条锈菌基因组DNA分离的氯化苄法和CTAB/SDS法进行了比较分析,认为后者无降解,质量高,可用于RAPD分析。试验对小麦条锈菌RAPD反应条件进行了优化,建立了标准化的小麦条锈菌RAPD反应体系,即10×Reaction Buffer 2.5 μL,MgCl2 2 mmol/L,dNTPs 0.15 mmol/L,模板DNA 40 ng,引物10 ng,Taq1U,ddH2O 15.76 μL。 |

Comparing two gDNA extration methods of the wheat stripe rust (Puccinia striiformis f.sp. tritici),the author regards the CTAB method is better than the PhCH2Cl method to search the special molecule markers.In order to facilitate communion,a standard RAPD system is also provided in this research. |
