为了从植物组织中快速提取核酸,从多种DNA提取方法中遴选出2种基本方法。通过优化基本方法的操作步骤、试剂用量与作用时间,建立了一种改良的DNA快速提取方法。在此方法中,用氯仿代替液氮或提取液进行植物组织研磨,之后加入提取液抽提DNA,并用氯仿-异戊醇反复抽担,可显著降低蛋白质含量;通过应用RNA酶可获得纯度较高的DNA。该方法的特点是在室温下即可进行简便操作,快速(提取核酸的过程仅需30 min),污染器皿少,实用性强,提取物产量高,纯度好。 |

Rapid method for extracting DNA from plant issues |

Abstract: |
Two methods were selected from various DNA isolated methods for extracting DNA from plant tissues rapidly.Through optimizing the steps,dosage of reagents and manipulating time,a reformative method is formed for extracting DNA rapidly.In our method,chloroform is replaced with liquid nitrogen,extracting reagent in which plant issues are skived was added, chloroform-isoamyl alcohol is used to extract DNA repeatedly,high yield of DNA is gained.Using RNase,varified DNA could be obtained.The trait of the method is simple to apply in room temperature,time-saving(30 min),less polluting,adaptable for extracting DNA from different plant materials and effective. |
