采用70 L环形通气搅拌生物反应器对长春花细胞的大规模悬浮培养进行了初步的研究,2次培养取得成功,培养周期15 d,最大PCV值达52%,生物量鲜重205.6 g/L。研究结果表明,“种子”细胞的接种时期和培养基中的蔗糖质量浓度对培养细胞在70 L反应器中的生长影响很大。从所做的2次成功培养中,将培养基的蔗糖质量浓度由20 g/L提高到25 g/L,按15%的接种量接种正处于生长指数初期的细胞,在70 L反应器中培养的长春花细胞长势最好,最大PCV值为52%,最终生物鲜重205.6 g/L。 |

Abstract: |
Catharanthus roseus cells was successfully cultivated in a 70 L stirred tank bioreactor.The batch culture period is 15 days,the maximal PCV is 52%,and the maximal biomass is 205.6 g/L in fresh.From our study,we draw the conclusion that the growth phase of "seeds" cell can greatly affects cell growth of cultures.High concentration of initial sucrose increased biomass accumulation.The suitable inoculum of Catharanthus roseus cells is 15% at initial exponential growing process. |
