河南省猪瘟流行毒的E2基因序列分析和基因分型

摘要:
采用RT-PCR和nPCR扩增出3株河南省近期(1999~2000年)猪瘟流行野毒的E2基因,分别克隆于PMD-18T载体,对其进行了核苷酸序列测定及氨基酸序列推导,同时进行了同源性比较及E2糖蛋白结构的分析。结果表明,所测3株HCVE2基因的长度均为1169bp。编码的氨基酸序列均包括完整信号肽序列和部分跨膜序列,并由384个氨基酸组成,3株流行毒的E2基因核酸序列同源性在99%以上,相应的氨基酸序列同源性也在99%以上;这3株流行毒与C-株兔脾组织毒(疫苗种毒)E2基因的核苷酸序列同源性在83.14%~83.23%,相应的氨基酸序列同源性在88.14%~88.68%。经遗传发生关系分析,C-株兔组织毒-C株细胞毒属于组群1(group1),河南省近期流行猪瘟毒属于组群2(group2),近期猪瘟流行毒与C-株疫苗毒的gp55蛋白之间存在一定的差异。

Sequence analysis and grouping of E2 gene of prevalent virulent strains of hog cholera virus in Henan Province
Abstract:
The E2 gene of three prevalent virulent strains of Hog Cholera Virus (HCV) from Henan,China were amplified by reverse transcription(RT) and the nested Polymerase Chain Reaction (nPCR).The amplifed E2 fragments of three HCV strains were all 1 169 bp in length by agarose gel electrophoresis.Three E2 fragments were cloned respectively into PMD-18T vector.1 169 bp cDNA fragments of three prevalent virulent E2 gene were sequenced and 384 residues amino acid sequences of E2 were deduced.The nucleotide homology among three prevalents virulents were over 99%;and that of amino acid were also 99%.The nucleotide identity of C-strain from spleen tissue of rabbit with that of three Chinese prevalent virulent strains were from 83.14% to 83.33%,and that of amino acid were from 88.14% to 88.68%.There revealed some differences in gp55 of C-strain and the 3 prevalent virulent strains.
