赤霉素荧光测定法改进研究

摘要:
研究了硫酸体积分数、硫酸用量、样品前处理及温度与时间对赤霉素测定结果的影响,对赤霉素的经典荧光测定法作了必要改进,并对荧光法与福林法的测定值进行了比较。结果表明:①改进荧光法与经典荧光法测定结果一致;②改进荧光法操作简便,不需冰浴及真空抽干设备;③节省试剂(50%~75%);④测定精确度和样品回收率高;⑤反应产物的荧光值具有一定的时间和温度稳定性;⑥该法适合于发酵液及固态产品中赤霉素的测定;⑦福林法选择性差,不能用于成分较为复杂的样品中赤霉素的测定。

Abstract:
The effects of sulphuric acid concentration,sulphuric acid quantity,sample pretreatment,temperature and time on gibberellin determination are studied.The classical fluorescence method for gibberellin determination is improved and compared with Folin method.The results indicate that:①The determination results of improved fluorescence method (IFM ) are consistent with the classical one.②Easily operated, and with no ice bath and vaccum pump equipment needed.③Reagent saving (50%~75% ).④High accuracy and high recoverys.⑤Fluorescence values of reactant are stable within 0~10 hs after reacting and 10℃~35℃.⑥IFM is suitable for the determination of gibberellin in fermented liquid and solid products.⑦Folin method can't be used for the determination of gibberellin in complicated samples without good selectivity.
