葡萄愈伤组织脯氨酸累积变异系的分离研究

摘要:
以羟脯氨酸(HYP)为选择剂,通过连续多步筛选法和间隙多步筛选法,从二倍体葡萄里扎马特(Rizamat)品种的能再生愈伤组织中,分离出了7个稳定的HYP抗性变异细胞无性系,连续多步选择法的选择效率为4. 24×10-5,间隙多步选择法的选择效率为7. 00×10-5.但连续多步选择法更有利于高脯氨酸累积变异系的选择。7个HYP抗性变异系都能稳定的累积脯氨酸,其游离脯氨酸的含量是对照的1. 79~3. 93倍。

Abstract:
Seven different lines resistant to HYP, designated as HR5,HR1, HR2, HR7, HR4, HR6 and HR3, have been recovered from diploid cultivar Rizamat callus by the continuous and interval multi-step selection system.The selection efficiencies are 4. 24×10-5 and 7. 00×10-5, respectively,while the continuous multi-step selection system is more effective than the interval multistep selection system.The proline accumulation in seven different lines resistant to HYP are stable and its free proline content of variant lines is 1. 79 to 3. 93 times higher than that of the control line.Plants are regenerated from both control and variant line callus.
