中华猕猴桃愈伤组织细胞的超低温伤害研究

摘要:
用5%二甲基亚砜+10%葡萄糖或5%二甲基亚砜+10%蔗糖作为冰冻保护剂,能有效减轻中华猕猴桃愈伤组织细胞的超低温伤害。以-1℃/min的速率降温至-60~-75℃时,对愈伤组织细胞的伤害作用最小;以大于160℃/min速率快速化冻,愈伤组织细胞的相对存活率最高。愈伤组织细胞伤害的主要温度区域为-20~-40℃;-60~-75℃为愈伤组织细胞投入液氮时的“安全温度区”,-60℃和-75℃分别为“临界上限温度”和“临界下限温度”。

The Injury of Actinidia chinensis Callus Cells in Super-low Temperature

Abstract:
Being as cryoprotectants,5% DMSO-10% glucuse or 5% DMSO-10% sucrose could greatly reduce the injury of Actinidia chinensis callus cells.The injury of callus cells was minimal when freezed at the rate of -l℃/min.to -60~-70℃. The survival rate of the callus cells was maximal when rewarmed at the rate of more than 160℃/min -20~-40℃ was the major temperature scope of the callus cell injury;-60~-75℃ was the security temperature scope when callus cells were immersed in liquid nitrogen;-60℃ and -75℃ were the upper and lower limits of critical temperatures respectively.
