早熟葡萄种子培养和胚培养

摘要:
结果表明:果实成熟前一周取样的种子培养后胚长1560μm,成熟期及成熟后一周取样的仅为953μm及830μm。培养基中蔗糖浓度在2—20%范围内浓度越低,效果越好。胚培养时培养基由加入GA,IA-A及IAA+KT均能促进胚的萌发,但对幼苗的生长有不同影响,在添加GA的培养基上幼苗生长正常。种子培养过程中胚不能自然萌发,种子培养后再结合胚培养可使胚萌发率达73.2%,不经种子培养而直接进行胚培养的,胚萌发率仅为5.6%。

Abstract:
The results indicated that san pling tine and sucrose concentration in the mediun were the two main factors affecting the embryo development during seed culture. The embryo growth in seed culture was increasingly inhibited when the sucrose concentration in the medium was increased from 2% to 20%. During embryo culture, when GA, IAA or IAA + KT were added to the medium, the embryo germination was improved but seedlings could grow normally only in the medium added with GA. Embryos did not germinate during seed culture. After seed culture, embryos were dissected and cultured on media and the germination rate reached up to 73%, compared to 5.6% for the embryos which had never undergone the seed culture.
