小鼠胚胎冷冻保存试验

摘要:
应用4~10周令的杂种 CB6/Han 和纯种 C57BL/Han 小鼠73只进行试验,共采胚胎523枚,将发育正常,形态完好的157枚胚胎低温冷冻保存。解冻后回收率为83.4%,其中45%(59枚)形态完好,(囊胚56%,桑椹胚20%)。将其培养24—48小时后,71.2%(42枚)继续发育到扩张囊胚(其中囊胚为76.5%,桑椹胚为37.5%)。应用3′6′—二乙酰基荧光素(FDA)染色进行胚胎质量评定,对胚胎无损害。

Abstract:
The experiment was conducted by using 73 mice of hybrid CB6/Han and pure bred C57BL/Han,aged 4-10 weeks,from which 523 embryos were collected.There were 157 normally developed and intact embryos to be preserved in freezing conditions.After unfreezing,the recovery percentage was 83.4%,of which 45% of embryos (59) remained intact (56% were blastocyst,and 20% morula).After 24-48 hours of culture,these embryos continued their development into expanding blastoysts,with the morulal being 37.5%,and the blastocyst 76.5%.3'6'-Fluorescein-Diacetyl (FDA) was used in staining,and no damage to the embryos was found.
