苹果茎顶的显微嫁接研究

本研究用金帅和斯巴丹(Spartan)两个苹果品种的茎顶在离体条件下分别嫁接在山定子、M7、M9和 M26的茎或根上,并获得成功。这种苹果显微嫁接方法尚未见报导。茎顶长度为0,2、0.4和0.7mm,砧木长度为0.5~1.2cm。嫁接成活率因接穗大小、嫁接方式而变化,其成活率范围在56.3%到81.3%之间。从获得无毒苗和具有较高的成活率两方面考虑,带有二片叶原基的茎顶作接穗是适当的。 嫁接七周后接穗干重因砧木不同而表现明显差异,矮砧上的接穗干物质积累量最少,半矮砧次之,乔化砧最多。作者认为,这可以作为一种鉴定砧木生长型的新途径,应进一步扩大砧木类型和品种的试验研究。

A Study of Micrografting of Apple Shoot Tips in Vitro
Abstract:
Micrografting of Golden Delicious and Spartan apples (Malus domestica Borkh) was achieved by placing a 0.2,0.4 or 0.7mm shoot tip on the 0.5-1.2cm stems or roots of Siberian crab seedling (Malus baccada L.),M7,M9 and M26.The percentage of successful graft unions in our study was increased from 56.3% to 81.3% with size of scions.The probability of obtaining the virus-free plantlets is inversely related to size of shoot tips and invitro micrografting desires more graft unions to develop into the plantlets.Our results showed that scion explants consisting of the dome and 2 leaf primordia were used in subsequent studies.The grafted explant unions developted into the planlets were affected by methods of micrograft. The dry weight of scions under in vitro conditions 7 weeks after grafting were affected by different vigor rootstocks significantly.In our study,the dry weight of apple explants on Sberian Crab (standardks rootstocks) was the highest,the second was M7 (semidwarfing rootstocks) and that on M9 or M25 (dwarfing rootatocks) was the lowest.This method could be used for the the determination of early dwarf apple rootstocks in breeding.
